Description

Revolade (Eltrombopag) is a thrombopoietin receptor agonist that stimulates platelet production by activating TPO receptors in the bone marrow. It is prescribed to increase platelet counts in patients with chronic immune thrombocytopenic purpura (ITP), aplastic anemia, or thrombocytopenia associated with chronic liver disease.

Indications

• Chronic immune thrombocytopenic purpura (ITP) in adults and children ≥ 1 year refractory to standard treatments.
• Severe aplastic anemia unresponsive to immunosuppressive therapy.
• Thrombocytopenia in chronic liver disease to reduce bleeding risk prior to invasive procedures.

Mechanism of action

Eltrombopag activates thrombopoietin receptors on megakaryocyte progenitors, stimulating platelet production and reducing bleeding episodes.

Composition

Each tablet contains:

• Active ingredient: Eltrombopag olamine equivalent to 25 mg Eltrombopag.
• Excipients: Mannitol, microcrystalline cellulose, croscarmellose sodium, magnesium stearate, hypromellose, titanium dioxide, ferric oxide yellow.

Dosage and administration

• Take orally with water.
• Administer at least 1 hour before or 2 hours after meals.
• Avoid dairy products and mineral supplements containing calcium, iron, magnesium, aluminum, or zinc near the dosing time.
• Dose is individualized by the treating physician.

Side effects

Common: headache, nausea, diarrhea, fatigue, elevated liver enzymes.
Rare: thrombocytosis, thromboembolic events, rash, myalgia.

Contraindications

• Hypersensitivity to Eltrombopag or any component.
• Severe hepatic impairment without medical monitoring.
• Pregnancy and breastfeeding unless clearly indicated.
• Pediatric patients under 1 year.

Precautions

• Regular monitoring of platelet count and liver function is required.
• Avoid concurrent use with cation-containing products.
• Discontinue gradually under physician supervision.

Storage

Store below 30 °C, in a dry place away from light.
Keep out of reach of children.
